腔腸素(55779-48-1,Coelenterazine) 是 apoaequorin 和 Renilla 熒光素酶的發(fā)光酶底物。Renilla 熒光素酶和底物 coelenterazine 被用作生物發(fā)光共振能量轉(zhuǎn)移 (BRET) 中檢測蛋白質(zhì)-蛋白質(zhì)相互作用的生物發(fā)光供體。腔腸素是一種超氧陰離子敏感化學(xué)發(fā)光鈣離子探針,其也用于過氧亞硝酸鹽的化學(xué)發(fā)光檢測。熒光 : λex 429 nm, λem 466 nm。

腔腸素(Coelenterazine),是海腎熒光素酶(Rluc)和Gaussia熒光素酶(Gluc)等多種熒光素酶的作用底物,腔腸素也是水母發(fā)光蛋白的輔助因子,應(yīng)用非常廣泛,可用于檢測活細胞中鈣離子濃度,基因報告分析,BRET(生物發(fā)光共振能量轉(zhuǎn)移)研究,ELISA,HTS,以及組織或細胞中ROS水平的化學(xué)發(fā)光檢測。

腔腸素(Coelenterazine)是自然界中資源最豐富的天然熒光素,是絕大多數(shù)海洋發(fā)光生物(超過75%)的光能貯存分子。腔腸素可作為許多熒光素酶的底物,比如海腎熒光素酶(Rluc),Gaussia分泌型熒光素酶(Gluc),以及包括水母發(fā)光蛋白(aequorin)和藪枝螅發(fā)光蛋白(Obelia)在內(nèi)的光蛋白(Photoproteins)。其發(fā)光原理是:以腔腸素為底物的熒光素酶在有分子氧的條件下,氧化腔腸素,產(chǎn)生高能量的中間產(chǎn)物,并在此過程中發(fā)射藍色光,峰值發(fā)射波長約為450~480nm。與甲蟲(或螢火蟲)熒光素/熒光素酶系統(tǒng)不同,腔腸素/熒光素酶系統(tǒng)不需要三磷酸腺苷(ATP),因此更便于體內(nèi)生物熒光的研究。因此,腔腸素常用作基于熒光分析的報告基因檢測以及活體動物檢測的發(fā)光底物。
腔腸素還能在酶非依賴性的氧化體系中自發(fā)熒光,細胞和組織內(nèi)的超氧陰離子和過氧化亞硝基陰離子能夠增強該自發(fā)光信號,因此其也可用來檢測細胞/組織內(nèi)活性氧(ROS)水平。?

腔腸素溶解性:溶于甲醇或者乙醇,不可溶于DMSO

腔腸素溶解特性:腔腸素及所有衍生物都溶于乙醇或者甲醇,切忌溶于DMSO。腔腸素的水溶性相當?shù)?,一般情況需用酸化的甲醇配制高濃度母液后再用水溶性緩沖液稀釋到需要的工作液,現(xiàn)配現(xiàn)用。
腔腸素母液的配制:稱取1mg腔腸素h粉末直接溶解于205μl酸化的甲醇溶液(100%甲醇中含有20μl/ml 3M或者6M HCl),配制成12 mM(~5mg/ml,5×)的腔腸素h儲存液。-20℃(最好-80℃)分裝避光保存,可能會有少量活性的喪失,但可保存4周。若存放在惰性氣體中可長期保存。

Coelenterazine produces luminescent signals following X-ray irradiation.
