RSV F蛋白高度保守,其功能為促進(jìn)病毒與宿主細(xì)胞的融合,也是中和抗體的重要靶點(diǎn)。雖然目前還不清楚F蛋白在感染過(guò)程中變構(gòu)是如何發(fā)生的,但是F蛋白的變構(gòu)現(xiàn)象(即從融合前pre-F構(gòu)象轉(zhuǎn)變?yōu)槿诤虾髉ost-F構(gòu)象)確實(shí)是自發(fā)發(fā)生的,也因此pre-F蛋白作為抗原的穩(wěn)定性很難保持,在FI-RSV候選疫苗中也主要展示了F蛋白融合后構(gòu)象(post-F)。然而,一些針對(duì)pre-F蛋白的穩(wěn)定突變已經(jīng)發(fā)現(xiàn),可以用來(lái)穩(wěn)定pre-F的構(gòu)象(圖1)。這些突變的發(fā)現(xiàn)也讓pre-F蛋白作為疫苗抗原成為了可能。

除F蛋白外,G蛋白是中和病毒表面的唯一其他靶點(diǎn)。G蛋白在病毒顆粒表面以低聚物的形式存在,并以單體形式從感染細(xì)胞中分泌出來(lái),對(duì)于病毒的附著至關(guān)重要。有證據(jù)表明單體可溶的G蛋白在病毒感染中起到誘餌的作用,幫助病毒逃避抗體反應(yīng)。然而G蛋白在RSV株間變異性較大,其表面結(jié)構(gòu)又知之甚少,因此不常用作疫苗抗原。
另一個(gè)可能的疫苗靶點(diǎn)是小疏水蛋白(SH),包含跨膜結(jié)構(gòu)域和胞外結(jié)構(gòu)域,與病毒在體內(nèi)的復(fù)制和炎癥激活方面起作用,可能能夠誘導(dǎo)抗體依賴的細(xì)胞介導(dǎo)細(xì)胞毒作用(ADCC),因此其胞外域已被用作抗原研究。
RSV的其他幾種非結(jié)構(gòu)蛋白(N蛋白,M2-1蛋白和M2-2蛋白)與誘導(dǎo)T細(xì)胞免疫息息相關(guān),因此在疫苗設(shè)計(jì)中也常作為疫苗抗原的靶點(diǎn),單獨(dú)或與其他抗原聯(lián)合使用。
