PROTAC (proteolysis targeting chimeras) 是一類異雙功能分子。PROTAC分子由三部分構(gòu)成,一端是結(jié)合需要降解的靶蛋白配體分子,另一端為結(jié)合E3泛素化連接酶的配體分子,中間則由特定的Linker連接器相連接。
ROTAC linker將E3泛素連接酶配體與靶蛋白配體連接起來(lái)。PEG是PROTAC開發(fā)中最常用的linker,據(jù)統(tǒng)計(jì),在已報(bào)道的PROTAC分子中有54%使用了PEG作為linker。
PEG linker在PROTAC中的作用:
1. PEG的引入可以增加PROTAC分子的水溶性,影響細(xì)胞滲透性,從而影響口服吸收。
2. 通過(guò)使用不同鏈長(zhǎng)的PEG可輕松實(shí)現(xiàn)對(duì) linker長(zhǎng)度系統(tǒng)性的改變,影響PROTAC分子的降解效率。
3. 使用各種雙官能化的PEG linker,能夠簡(jiǎn)單、快速地組裝具有不同連接基團(tuán)的分子,加速篩選得到有效的降解結(jié)構(gòu)。
