FITC標記葡聚糖是由熒光素異硫氰酸酯(5-異構(gòu)體)與相應的葡聚糖偶聯(lián)合成的。FITC標記葡聚糖是黃色至橙色粉末,易溶于水或鹽溶液,呈黃色溶液。該產(chǎn)品也可以溶于DMSO,甲酰胺和其它極性有機溶劑,但在低級脂肪醇,丙酮,氯仿和DMF中基本不溶。葡聚糖是來源于腸系膜白串珠菌B-512F的分支多糖,本質(zhì)上是一種α-(1-6)鏈接的線性葡萄糖鏈。分支比例大約是5%。熒光素是通過穩(wěn)定的硫代氨甲酰鍵鏈接的,標記過程不會引起任何葡聚糖的解聚。FITC標記葡聚糖,每個葡萄糖單元對應0.002到0.008 mol的FITC。 這些低取代度使葡聚糖電荷極小,這也是滲透性研究最基本的要求。

Dextran is a water-soluble polysaccharide extracted from the metabolites of bacteria (e.g. Candida albicans). It is composed of D-glucose linked by 1 → 6 bonds and linked by glycosylation at branch points 1 → 2, 1 → 3 and 1 → 4. According to different sources, various branched glucans with different average relative molecular weights can be obtained. At first, glucan was used as a substitute for plasma. Because of its biodegradability, non-specific cell adhesion, resistance to protein adsorption, low price and easy structural modification, it has become a hotspot in the research of carrier materials.
