巰基官能化PEG是一種單官能PEG衍生物可以用來修飾蛋白質(zhì)、多肽以及其它材料或者小分子和生物材料。具有無反應(yīng)性的巰基、半胱氨酸,馬來酰亞胺和巰基在PH6.5-7.5很容易形成穩(wěn)定的硫醚鍵。硫醇基團對金表面具有很高的親和性,用于金納米粒子或金膜修飾。聚乙二醇修飾后可提高溶解度和穩(wěn)定性,降低蛋白質(zhì)和肽的免疫原性,它也還能能抑制帶電分子在修飾表面的非特異性結(jié)合。通過S-S二硫鍵輕松空氣氧化形成二聚體,可通過還原劑逆轉(zhuǎn)。對于可逆聚乙二醇化是有用的。

Thiol functionalized peg is a monofunctional peg derivative that can be used to modify proteins, polypeptides and other materials or small molecules and biomaterials. Non reactive sulfhydryl, cysteine, maleimide and sulfhydryl groups can easily form stable thioether bonds at pH 6.5-7.5. Thiol groups have high affinity for gold surface and are used to modify gold nanoparticles or gold films. PEG modification can improve the solubility and stability, reduce the immunogenicity of proteins and peptides, and inhibit the non-specific binding of charged molecules on the modified surface. Dimer is easily formed by air oxidation through S-S disulfide bond, which can be reversed by reducing agent. Useful for reversible PEGylation.
