1.百螢D-熒光素鉀鹽工作原理
? ? ? ?D-熒光素是流行的多功能生物發(fā)光底物。螢火蟲熒光素酶/熒光素生物發(fā)光系統(tǒng)存在于螢火蟲(Photinus pyralis)和其他幾種甲蟲中。熒光素酶通過二氧雜環(huán)丁酮中間體氧化ATP進(jìn)行活化熒光素。螢火蟲熒光素酶通過熒光素的ATP依賴性氧化產(chǎn)生光。來自該反應(yīng)的560nm化學(xué)發(fā)光在數(shù)秒內(nèi)達(dá)到峰值,當(dāng)熒光素和ATP過量存在時(shí),光輸出與熒光素酶活性成比例。螢火蟲熒光素酶長期以來與抗體結(jié)合,并在熒光素作為檢測底物的免疫測定中用作標(biāo)記物。與HRP和堿性磷酸酶相比,熒光素酶對(duì)化學(xué)修飾的耐受性較差。該酶的一個(gè)特別優(yōu)點(diǎn)是除了其高靈敏度之外,在哺乳動(dòng)物組織中存在低內(nèi)源熒光素酶活性。熒光素酶的另一個(gè)重要用途是衛(wèi)生監(jiān)測領(lǐng)域,熒光素酶/熒光素系統(tǒng)可用于檢測污染,因?yàn)榇嬖谟谒猩矬w中的ATP需要發(fā)光,這種ATP生物發(fā)光的主要應(yīng)用是通過測試食品加工廠中的表面來確定質(zhì)量,以確定是否存在設(shè)備或產(chǎn)品的污染。
