DOTA-PSMA is a strong and effective inhibitor of Prostate specific membrane antigen (PSMA), with a Ki value of 0.37 nM. DOTA-PSMA is composed of three components: the pharmacodynamic group Glutamate-urea-Lysine, the chelating agent DOTA (capable of binding 68Ga or 177Lu), and the linker connecting the two entities.
DOTA is the gold standard that complicates most radiometers. It is important to find the right combination between chelating agent and biological carrier. Many factors, such as size, geometry, lipophilicity and the charge of metal complexes, can affect the pharmacokinetics.
