2',3'-環(huán)鳥苷單磷酸-腺苷單磷酸(環(huán)GMP-AMP,cGAMP,環(huán)[G(2',5')pA(3',5'')p])是在后生動物中發(fā)現(xiàn)的第一個環(huán)狀二核苷酸.2',3'-cGAMP也稱為由于在鳥苷和腺苷。2',3'-循環(huán)GAMP是一種新的先天免疫第二信使調(diào)節(jié)I型干擾素(IFN)的產(chǎn)生1-6。由cGAS(cGAMP合成酶)在哺乳動物細胞中產(chǎn)生作為對細胞質(zhì)中雙鏈DNA與cGAS結(jié)合的反應,cGAMP與干擾素基因(STING)。隨后STING誘導TBK1-IRF3依賴性IFN-β的產(chǎn)生。

DetectX?2',3'-循環(huán)GAMP(cGAMP)免疫測定試劑盒設(shè)計用于定量測量存在于裂解的細胞和組織、EDTA血漿和組織培養(yǎng)基樣品中的2',3'-cGAMP。在進行此測定之前,請閱讀完整的試劑盒說明書。提供2',3'-cGAMP標準生成用于測定的標準曲線,并且應當從標準曲線讀取所有樣品。清晰提供了用抗體包被以捕獲兔IgG的微量滴定板。標準品或稀釋樣品被移到井中。將2',3'-cGAMP過氧化物酶綴合物添加到標準品和樣品中在井里。通過將兔多克隆抗體加入2',3'-cGAMP來引發(fā)結(jié)合反應每個井。培養(yǎng)2小時后,洗滌板并加入基質(zhì)?;|(zhì)反應用結(jié)合的2',3'-cGAMP過氧化物酶綴合物,短時間孵育后停止反應并且在能夠測量450的微量滴定板讀取器中檢測產(chǎn)生的顏色的強度nm波長。在制作合適的樣品后,計算樣品中2',3'-cGAMP的濃度使用大多數(shù)讀板器可用的軟件對樣品稀釋度進行校正。
