摘要
B細(xì)胞生物學(xué)的多個(gè)檢查點(diǎn)中,通過B細(xì)胞受體(BCR)傳導(dǎo)的信號(hào)起著關(guān)鍵作用。比如,BCR在骨髓(BM)的早期發(fā)育過程中扮演守門人的角色;在克隆刪除和受體編輯等觸發(fā)機(jī)制中也不可替代,能消除自身反應(yīng)性B細(xì)胞;外周循環(huán)時(shí),最重要的一項(xiàng)功能是作為受體識(shí)別各種細(xì)胞外抗原,包括應(yīng)對(duì)細(xì)菌和病毒感染,來增強(qiáng)宿主防御能力(BCR對(duì)抗原的識(shí)別是獲得T細(xì)胞幫助幼稚B細(xì)胞向漿細(xì)胞、生發(fā)中心B細(xì)胞和記憶B細(xì)胞分化的第一步);此外,類似于在B細(xì)胞發(fā)育早期發(fā)揮的作用,BCR信號(hào)在防止自身反應(yīng)性B細(xì)胞的異常激活中也意義重大,因?yàn)楫惓<せ羁烧T導(dǎo)次級(jí)淋巴器官相關(guān)的自身免疫生成。BCR對(duì)于B細(xì)胞正確地激發(fā)免疫反應(yīng)必不可少,通過BCR的信號(hào)也受到細(xì)胞內(nèi)正負(fù)調(diào)控因子的嚴(yán)格控制。在這一章中,根據(jù)最新的研究,我們對(duì)BCR信號(hào)的激活和抑制機(jī)制進(jìn)行了綜述。
