百螢 鈣藍(lán)素AM,細(xì)胞滲透型鈣黃綠素
? ? ? 鈣藍(lán)素,AM是美國AAT Bioquest生產(chǎn)的細(xì)胞滲透性鈣黃綠素,它是鈣黃綠素的細(xì)胞滲透型。進(jìn)入活細(xì)胞后,弱熒光鈣黃綠素AM被水解成鈣黃綠素,其激發(fā)/發(fā)射大值與DAPI,Hoechst和AMCA相似。與典型的綠色和紅色熒光團(如FITC,TMR和德克薩斯紅)的這種特殊光譜分離為多路復(fù)用實驗提供了額外的選擇。因為鈣黃綠素AM本身是熒光的,所以可能需要適當(dāng)?shù)倪^濾器組和額外的洗滌步驟以使背景熒光小化。我們的鈣測定緩沖液可用于有效洗滌細(xì)胞外鈣黃綠素。

3.?百螢 鈣藍(lán)素AM實驗方案
操作步驟
(1)準(zhǔn)備HHBS緩沖液,10%Pluronic?F-127溶液和25 mM Probenecid溶液。
(2)在高質(zhì)量無水DMSO中制備2 mM至5 mM Calcein Blue,AM * CAS 168482-84-6 *儲備液。
(2.1)卡介素藍(lán)量,AM * CAS 168482-84-6 *使用量:1毫克
(2.2)所需濃度:2 mM
(2.3)在合適的容器中,將1mg鈣黃綠素,AM * CAS 168482-84-6 *與1074.32μL無水DMSO混合。
?(3)用含有10μM鈣黃綠素的HHBS制備2X工作溶液,AM * CAS 168482-84-6 * 4,0.08%Pluronic?F-127和2 mM丙磺舒。
(3.1)Calcein Blue的終孔內(nèi)濃度,AM * CAS 168482-84-6 *:5μM
(3.2)Pluronic?F-127的終孔內(nèi)濃度:0.04%
(3.3)終孔內(nèi)濃度的丙磺舒:1mM
(3.4)在合適的容器中混合16μL鈣黃綠素,AM * CAS 168482-84-6 *,25.6μL10%Pluronic?F-127和256μL25mM Probenecid。然后,添加HHBS或您選擇的緩沖液,直到體積為3.2 mL。
注意:對于大多數(shù)細(xì)胞系,我們建議終濃度為Calcein Blue,AM * CAS 168482-84-6 *為4至5μM。
注意:推薦的Pluronic F-127井濃度終為0.02%至0.04%。
注意:推薦的終濃度為1至2.5 mM的Probenecid。
?(4)將100μL染料工作溶液加入已經(jīng)含有100μL培養(yǎng)基的所需孔中。
(4.1)此步驟將染料工作溶液從2X稀釋至1X,并將每種組分的終濃度調(diào)整為以下:5μM鈣黃綠素,AM * CAS 168482-84-6 *,0.04%Pluronic?F-127,1 mM丙磺舒。
?(5)孵育染料
(5.1)將染料加載板在細(xì)胞培養(yǎng)箱中孵育20-60分鐘。
(5.2)將染料加載板在室溫下孵育30分鐘。
?(6)用1.0 mM Probenecid準(zhǔn)備HHBS緩沖液(或您選擇的緩沖液)。
在合適的容器中加入160μL的25mM丙磺舒。 接下來,添加HHBS或您選擇的緩沖液,直到體積為4 mL。
?(7)用HHBS緩沖液或您選擇的緩沖液替換染料工作溶液,使用1.0 mM Probenecid。
(7.1)首先,從所需孔中除去200μL染料工作溶液和培養(yǎng)基。
(7.2)在相同的孔中加入200μL含有1.0mM丙磺舒的HHBS(或您選擇的緩沖液)。
?(8)運行實驗
(8.1)為您的樣品添加所需的處理。
(8.2)以Ex / Em = 360/445 nm運行實驗。
