呼吸道合胞病毒(Respiratory Syncytial Virus, RSV)作為引起急性下呼吸道感染(ALRTIs)的主要原因之一,每年導(dǎo)致全球約3.31億人發(fā)生呼吸道感染。然而,就是這樣一個常見且早已被發(fā)現(xiàn)的呼吸道病毒,其疫苗的研發(fā)卻十分坎坷,以至于至今仍無一款成熟的RSV疫苗問世。
此前的數(shù)十年間,科學(xué)家們不曾停止對于RSV疫苗的研究與開發(fā),但均以失敗告終。究其原因,可能是因?yàn)镽SV病毒結(jié)構(gòu)的特殊性。病毒表面的融合蛋白(fusion protein , F)和粘附蛋白(attachment protein, G)因在病毒入侵過程中的關(guān)鍵作用成為病毒研究的理想靶點(diǎn),而其中G蛋白變異較多相對不穩(wěn)定,F(xiàn)蛋白也存在Pre到Post構(gòu)象轉(zhuǎn)變影響其穩(wěn)定性。

為了克服這一障礙,科學(xué)家們對RSV F蛋白的蛋白結(jié)構(gòu)及其抗體進(jìn)行了研究,發(fā)現(xiàn)該蛋白表面至少存在7個完全不同的抗原表位,并成功篩選到可以與不同表位結(jié)合的抗體,分別為φ site_D25/5C4/AM22,I site_131-2A/2F,II site_motavizumab/palivizumab,III site_MPE8,IV site_101F/mAb19,V site_hRSV90。其中識別IV表位的101F、識別II表位的palivizumab和識別I表位的131-2A抗體可以同時識別F蛋白的pre和post構(gòu)象,識別φ表位的D25抗體只與Pre構(gòu)象的F蛋白發(fā)生結(jié)合,識別III表位的MPE8抗體則偏好識別Pre構(gòu)象,與Post構(gòu)象具有較弱的結(jié)合活性。2010年,Kwakkenbos研究團(tuán)隊(duì)篩選獲得單克隆抗體AM14,其識別的抗原表位位于F-pre蛋白近赤道區(qū)域,橫跨兩個單體,可特異性識別F-pre三聚體結(jié)構(gòu),且被證明與此前中和效果最佳的D25抗體具有相似的中和活性和更快的動力學(xué)性質(zhì)。
