G,F(xiàn)蛋白是2個(gè)重要的膜表面糖蛋白,Levine等[9–10]通過G蛋白與F蛋白單克隆抗體反應(yīng)發(fā)現(xiàn)2種蛋白分別促進(jìn)病毒與細(xì)胞受體黏附并介導(dǎo)細(xì)胞融合,同時(shí)兩者都是產(chǎn)生保護(hù)性抗體的靶點(diǎn)。
G蛋白是由約298個(gè)氨基酸(aa)組成的高度糖基化膜蛋白,以分泌型(Gs)和膜錨定型(Gm)兩種方式進(jìn)行表達(dá),其中Gs與抗體中和有關(guān),Gm與病毒附著有關(guān)。G蛋白作為中和抗體的靶點(diǎn)之一,不僅是重要的保護(hù)性抗原而且輔助F蛋白進(jìn)行細(xì)胞融合。G蛋白總體構(gòu)架類似于粘蛋白,但是相對于其他副黏病毒粘蛋白,缺少了血細(xì)胞凝集素與神經(jīng)氨酸酶的活性[9]。G蛋白主要含有胞漿區(qū)(aa1~aa37)、跨膜區(qū)(aa38~aa67)和胞外區(qū)(aa68~aa298),胞外區(qū)域由2個(gè)粘蛋白區(qū)與13個(gè)aa組成的中心保守區(qū)組成,此保守區(qū)域含有CX3C模序(182aa~186aa)以及中和抗體的結(jié)合位點(diǎn)[6–7,?10]。CX3C模序與CX3CR1受體結(jié)合,利于病毒吸附,擴(kuò)大感染范圍。目前,國際上普遍依據(jù)G蛋白第二粘蛋白區(qū)的第二高變區(qū)核苷酸序列對HRSV進(jìn)行分型[9–12]??乖宰儺愇挥贕蛋白的粘蛋白結(jié)構(gòu)域中,N和O糖基化可促進(jìn)蛋白成熟并與免疫逃逸機(jī)制增強(qiáng)有關(guān)。G蛋白高度可變,在亞群A和B之間氨基酸序列上有50%差異。
F蛋白是由574個(gè)aa組成的一類融合蛋白,主要負(fù)責(zé)病毒與宿主細(xì)胞融合,是中和抗體結(jié)合位點(diǎn)以及疫苗開發(fā)的主要目標(biāo)蛋白,同時(shí)F蛋白也是人類白細(xì)胞抗原(HLA)限制性細(xì)胞毒性T細(xì)胞(CTL)識別的靶點(diǎn)。研究發(fā)現(xiàn),A亞型與B亞型F蛋白基因276位點(diǎn)會發(fā)生基因突變,導(dǎo)致天冬氨酸突變?yōu)榻z氨酸,并認(rèn)為是帕麗珠單抗(PZ)的非抗性突變。
