? ?張可博士的研究方向是環(huán)境生物化學(xué),以應(yīng)對自然和工程生物污染物帶來的環(huán)境挑戰(zhàn)。他的最終研究目標(biāo)是預(yù)測生物污染物在環(huán)境系統(tǒng)中的行為,并優(yōu)化策略以將其對人類和生態(tài)系統(tǒng)健康造成的風(fēng)險降至最低。他在圣路易斯華盛頓大學(xué)獲得了博士學(xué)位,研究重點(diǎn)是基于 RNA 干擾的生物農(nóng)藥的環(huán)境歸趨。目前,他是密歇根大學(xué)的博士后研究員,主要研究流感病毒在室內(nèi)環(huán)境中的歸趨。
報(bào)告摘要
? In recent years, scientists have developed RNA interference crops that express double-stranded RNA (dsRNA) biopesticides in their tissues. While these genetically modified crops have shown to be effective in controlling pests, they also release dsRNA biopesticides to receiving environments and pose potential ecological risks to non-target organisms depending on the environmental concentration of dsRNA biopesticides. My work aimed to enable the accurate ecological risk assessments of dsRNA biopesticides. Firstly, I developed a new method using quantitative reverse transcription polymerase chain reaction (RT-qPCR) to measure dsRNA in agricultural soils at environmentally relevant concentrations. Secondly, I characterized the chemical stability of dsRNA in aqueous solutions by examining the impacts of the duplex structure of dsRNA on alkaline hydrolysis. Finally, I elucidated a novel abiotic reaction by which RNA, but not DNA, degraded upon adsorption to minerals in soils and sediments under environmentally relevant physicochemical conditions. These studies advance our understanding of the fate of dsRNA biopesticides in receiving environments and contribute to the ecological risk assessment of dsRNA biopesticides.
